Tau protein in the brain may harm memory partly by breaking up the slow waves of deep sleep

Most of us have noticed that memory feels sharper after a good night’s sleep. Memory also tends to slip with age, and more so in people developing Alzheimer’s disease. A study of young and older adults, combining overnight sleep recordings with brain scans, suggests one way sleep and that memory loss may be tied together.

The link runs through the slow electrical waves of deep sleep. In older adults with more of the protein tau in the front of the brain, those waves stopped behaving as a coordinated sweep and became isolated, stalled events. People whose slow waves stalled more also held on to less of what they had learned the night before.

What is tau, and why does it matter for memory?

Tau is a protein found inside nerve cells. In Alzheimer’s disease it clumps into tangles that build up and spread through the brain. The authors open with a blunt observation: memory markedly declines with age, more so in people with Alzheimer’s, yet the mechanisms are still not resolved. Tau deposits are closely tied to memory loss, but it has been less clear whether tau does its damage directly or partly through something else.

What are the slow waves of deep sleep?

During the deepest stage of non-REM sleep, the brain’s electrical activity settles into large, slow rhythms. These slow waves can be recorded with electrodes on the scalp using a test called electroencephalography, or EEG. They are thought to help the brain move new memories into longer-term storage overnight.

A healthy slow wave is not a single local flicker. The figures in the paper show slow waves traveling across the brain in both young and older adults’ recordings. Many regions join in together, and the wave sweeps across the cortex like a ripple moving over a pond.

Writing in Nature Neuroscience, the team combined three kinds of measurement. They recorded participants’ brain activity overnight. They used positron emission tomography (PET), a scan that can show where tau has built up in the living brain. And they measured how well people retained memories across the night.

In older adults, frontal lobe tau was associated with slow waves that lost their unity and failed to travel across the cortex. The researchers call these lonely, nontraveling slow waves. The problem with slow-wave travel was restricted to the frontal lobe and predicted by how much tau had accumulated there.

Those stalled waves mattered for memory. The paper reports that slow-wave travel linked the effects of frontal tau to overnight memory consolidation, meaning the loss of traveling waves appeared to sit between the tau and the weaker memory.

It did, and that is one of the stronger points of the study. The team repeated and extended the analysis in an independent clinical group, this time using markers of Alzheimer’s disease measured in cerebrospinal fluid, the liquid that surrounds the brain and spinal cord, rather than PET scans.

Getting a similar answer with a different group of people and a different way of measuring the disease makes it less likely the first result was a fluke of one sample or one scanning method.

Does this mean poor sleep causes memory loss?

Not on this evidence. The study shows an association: more tau went with more stalled slow waves, and more stalled slow waves went with weaker overnight memory. It cannot show which came first, and the full details of the sample and its limitations sit behind a paywall that was not reviewed for this article.

The authors’ conclusion is carefully worded. They argue that tau-associated memory problems are not wholly direct, and are partly linked to these consequential lonely waves. That leaves room for tau to harm memory in other ways too.

Could sleep be a target for protecting memory?

This is where the finding becomes interesting beyond the lab. If part of tau’s effect on memory runs through disrupted sleep waves, then the sleep waves themselves become something that could, in principle, be measured and perhaps influenced, although this study did not test any way of doing that.

For now, nothing about this study changes what anyone should do. Anyone noticing changes in their memory, or in a relative’s, should talk to a health professional. What the research adds is a clearer sense that sleep may be part of the story of memory loss in aging, not just a bystander to it.
